Effect of seasonal variation in trophic conditions and the gametogenic cycle on d13C and d15N levels of diploid and triploid Pacific oysters Crassostrea gigas ArchiMer

Carbon and nitrogen stable isotopes were investigated in separate organs of diploid and sterile triploid Pacific oysters Crassostrea gigas for 13 mo, together with changes in chemical and isotope composition of suspended matter sampled from an intertidal mudflat within Marennes-Oléron Bay, France. Particulate organic matter (POM) was a mixture of pelagic and benthic material with a predominance of neritic phytoplankton in spring, and resuspended microphytobenthos in summer and autumn. A remarkable shift of +30/00 in d13C was reflected in both diploids and triploids from spring to summer, and further temporal differences were observed amongst their tissues. Episodes de mortalité massive de moules bleues observés en 2014 dans les Pertuis charentais ArchiMer

Mass mortality outbreaks (90-100%) in both juvenile and adult blue mussels were reported in 2014 along the Pertuis Charentais area of the French Atlantic coast. A series of studies were carried out to learn more about the phenomenon through detecting pathogens, describing environmental conditions and establishing the physiological status of the animals. Bacteria identif ied as belonging to the V. splendidus species were detected in dying mussels and seem able to cause mortality in blue mussels in the laboratory. Unusual environmental conditions (large amounts of fresh water, re-suspension of sediments during a series of storms, and a high renewal rate of specific water masses in Brittany’s Pertuis area) may be partially responsible for sparking the phenomen

La décontamination bactériologique des huitres Crassostrea gigas a été étudiée expérimentalement dans des bassins dégorgeoirs de 13 m3 de conception proche de la configuration normale d'une station de purification française. Les huîtres contaminées artificiellement ont été placées dans des conditions dites "naturelles" d'épuration. Il n'a pas été utilisé de procédé physique ou chimique de purification de l'eau. The germs decreasing of artificially contaminated oysters Crassostrea gigas has been studied in experimental tanks similar to a shellfish depuration plant, without any chemical or physical water treatment. The oyster were treated by three different water management pattern, compared to a control tank and the germs decreasing studied by MPN method of fecal Coliforms and Streptococus. As a result: 1) it appaears somewhat difficult to conclude in this first attempt about the most efficient treatment. 2) the most important factor in shellfish depuration working is obviously the physiological activity (and stage) of the animals.

Some water samplings and bacteriological analyses carried out in the centre of the basin (pointe du Chapus) in 1979 (Heral and Prou 1980) show a good average water quality. In the summer, a pollution linked to tourist activities in the region shows an increase in the number of faecal coliforms.

In order to better understand the chronic summer mortality striking the Marennes Oléron Bay since 1988, a research program has been developed since 1994 in association with the professional sector. This study comprises the monitoring, from March to October 1997, of a homogeneous oyster population cultivated on-bottom and off-bottom on 15 working sites. One of these sites is equipped with a multiparameter probe and a valvometer. Measures are taken on an on-going basis on the working site.

The Marennes-Oléron is a well known cupped oyster (Crassostrea gigas) and mussel (Mytilus edulis) reproduction area. Every year, a monitoring of the reproduction of these two species is carried out on behalf of the Regional Shellfish Culture Section of Marennes-Oléron. This monitoring aims to determine the periods of larval emission in order to let the professionals know about the best collection times This study presents the main results obtained in 1999 and compares them with those obtained in 1997 and 1998. The year 1999 showed good results as far as larval emission and development are concerned in both oysters and mussels. The comparison of the three consecutive years shows the disparity of larval evolutions.

Massive abnormal mortalities of flat oysters Crassostrea during the summer period are nothing new, and such episodes have been described in scientific literature for about forty years. Numerous studies demonstrate that there is not one single identified cause to this phenomenon and that it is, on the contrary, a combination of different parameters. This program proposes a multidisciplinary approach to the problem of summer mortality of C. gigas in which the working hypothesis is that these mortalities result from the interaction of three parameters: environment, host and pathogens. L'aménagement du fleuve Charente constitue un enjeu particulièrement important tant au niveau des activités de la bande côtière que pour le bassin versant. Au niveau du littoral charentais, et en particulier pour la conchyliculture, le fleuve Charente représente le principal apport d'eaux douces, et donc d 'éléments nutritifs, essentiels au développement de la production primaire et des larves d'huîtres. Ainsi, les apports d'eau douce sont importants pour le développement du phytoplancton (printemps, été, automne), le développement des larves d'huîtres en période estivale qui requiert une dessalure, et la qualité des huîtres en fin d'année. Experimental infection of Pacific oyster Crassostrea gigas spat by ostreid herpesvirus 1: demonstration of oyster spat susceptibility ArchiMer

In 2008 and 2009, acute mortalities occurred in France among Pacific cupped oyster, Crassostrea gigas, spat. Experimental ostreid herpesvirus 1 infection of the Pacific oyster Crassostrea gigas: Kinetics of virus DNA detection by q-PCR in seawater and in oyster samples ArchiMer

Herpes- and herpes-like viruses are known to infect a wide range of bivalve mollusc species throughout the world. Abnormal summer mortalities associated to the detection of ostreid herpesvirus 1 (OsHV-1) have been currently reported in France among larvae and spat of the Pacific cupped oyster Crassostrea gigas. In the present work, we have developed an experimental protocol of horizontal transmission based on the cohabitation between healthy and experimentally infected oysters. Through a cohabitation trial, the kinetics of OsHV-1 detection in different oyster organs and seawater samples were investigated and characterized for the first time using real time quantitative PCR.

Viral entry mechanisms of herpesviruses constitute a highly complex process which implicates several viral glycoproteins and different receptors on the host cell surfaces. This initial infection stage was currently undescribed for Ostreid herpes virus 1 (OsHV-1), a herpesvirus infecting bivalves including the Pacific oyster, Crassostrea gigas. To identify OsHV-1 glyproteins implicated in the attachment of the virus to oyster cells, three viral putative membrane proteins, encoded by ORF 25, 41, and 72, were selected and polyclonal antibodies against these targets were used to explore first interactions between the virus and host cells. Extracellular products (ECPs) of the French V. tubiashii strain 07/118 T2 were previously reported to be toxic for the Pacific oyster Crassostrea gigas. In this study we now assessed host cellular immune responses and bacterial potential effectors by which these ECPs can be associated with host damages. The adhesion capacity (28% inhibition) and phagocytosis ability (56% inhibition) of oyster hemocytes were the main functions affected following in vitro contact between hemocytes and V. tubiashii ECPs. First detection of OsHV-1 in the cephalopod Octopus vulgaris. Is the octopus a dead-end for OsHV-1? ArchiMer

The ostreid herpes virus (OsHV-1), associated with massive mortalities in the bivalve Crassostrea gigas, was detected for the first time in the cephalopod Octopus vulgaris. Wild adult animals from a natural breeding area in Spain showed an overall prevalence of detection of 87.5% between 2010 and 2015 suggesting an environmental source of viral material uptake. Overall positive PCR detections were significantly higher in adult animals (p = 0.031) compared to newly hatched paralarvae (62%). Prevalence in embryos reached 65%. Since 2008, massive mortality events have been reported among C. gigas in Europe in relation to the detection of a variant of OsHV-1, called μVar. Monitoring hydrology is an essential part of studying an ecosystem such as the Marennes-Oléron Bay. Since 1977, bimonthly samples have been taken from five stations in the bay. These samples provide important information on physical characteristics (temperature, oxygen, salinity), nutrients (NH4, NO3, NO2, SiO3, PO4) and particulate matter composition (seston, chlorophyll a and pheophytin).
